Isolation and Characterization of Receptor Sequences Figure 1A diagrams four recombinant inserts recovered from cDNA libraries constructed from J2. 17 cell RNA; J2. 17 is a derivative of the rat hepatoma cell line, HTC (Ringold et al., 1979). As shown, the four overlapping clones together encompass virtually the entire length of the largest (approximately 6.5 kb) glucocorticoid receptor transcript in J2. 17 (Figures 1A and 1B). The initial clones (which include pRM18 and pRM9) were obtained by partial purification of receptor mRNA by polysome immunoenrichment, cDNA construction, and differential screening with probes produced from immunoenriched RNA and from total RNA (Miesfeld et al., 1984). Using DNA sequence information from these clones, a second cDNA library was produced by specific oligonucleotide priming of total J2. 17 RNA; screening this library with pRM9 sequences yielded pTS3 …
